چکیده

Note Rho-like GTPases belong to the Ras superfamily and have crucial roles in many cellular processes, such as regulation of the actin cytoskeleton, cell migration, cell cycle progression, gene transcription and cell adhesion. Rho-like GTPases cycle between a GDP-bound inactive form and a GTP-bound active form and activation is catalysed by guanine nucleotide exchange factors (GNEFs) (Minard et al., 2004). The GNEF Tiam1 has been identified as an invasionand metastasis-inducing gene in a murine T-lymphoma cell line and specifically activates the Rho-like GTPase Rac. Tiam1/Rac signaling controls the establishment and maintenance of E-cadherin-based cell-cell adhesions and loss of Tiam1 leads to epithelialmesenchymal transition (EMT). Consequently, Tiam1/Rac signaling affects cell migration, invasion and tumor metastasis, but to some extent these effects seem to be cell typeand tumor type-dependent (Ellenbroek et al., 2007). Moreover, Tiam1 and Rac have been implicated in oncogenic transformation of cells.

Tiam 1 Effects on Invasive Capacity of Gastric Cancer Cells in vitro and Underlying Mechanisms

Despite advances in surgical treatment, chemotherapy and radiotherapy, gastric cancer remains a major global health burden. The most recent estimates show that it is the fourth most common cancer and the second most common cause of cancer deaths worldwide (Lee et al., 2012; Mahar et al., 2012). Most victims of patients with gastric cancer died as a result of complications associated with recurr...

Focal adhesion kinase regulates syndecan-2-mediated tumorigenic activity of HT1080 fibrosarcoma cells.

Expression of syndecan-2, a transmembrane heparan sulfate proteoglycan, is crucial for the tumorigenic activity in colon carcinoma cells. However, despite the high-level expression of syndecan-2 in mesenchymal cells, few studies have addressed the function of syndecan-2 in sarcoma cells. In HT1080 fibrosarcoma cells, we found that syndecan-2 regulated migration, invasion into Matrigel, and anch...

The Rac activator Tiam 1 is required for 3 1 - mediated laminin - 5 deposition , cell spreading , and cell migration Irene

he Rho-like guanosine triphosphatase Rac1 regulates various signaling pathways, including integrin-mediated adhesion and migration of cells. However, the mechanisms by which integrins signal toward Rac are poorly understood. We show that the Rac-specific guanine nucleotide exchange factor Tiam1 (T-lymphoma invasion and metastasis 1) is required for the integrin-mediated laminin (LN)-5 depositio...

Recruitment of Tiam1 to Semaphorin 4D Activates Rac and Enhances Proliferation, Invasion, and Metastasis in Oral Squamous Cell Carcinoma

The semaphorins and the plexins are a family of large, cysteine-rich proteins originally identified as regulators of axon growth and lymphocyte activation that are now known to provide motility and positional information for a number of cell and tissue types. For example, our group and others have shown that some malignancies over express Semaphorin 4D (S4D), which acts through its receptor Ple...
